实验方法原理

产品仅用于科研此技术利用PCR技术能够在体外进行有效的基因重组，而且不需要内切酶消化和连接酶处理，可利用这一技术很快获得其它依靠限制性内切酶消化的方法难以得到的产物。重叠延伸PCR技术成功的关键是重叠互补引物的设计。重叠延伸PCR在基因的定点突变、融合基因的构建、长片段基因的合成、基因敲除以及目的基因的扩增等方面有其广泛而独特的应用。

实验步骤

1.  以引物a/b进行pcr1。

2.  以引物c/d进行pcr2。

3.  引物b/c中引入了需要的限制性位点。

4.  混合二者pcr产物，混合以前*回收一下。

5.  进行pcr延伸反应，只有右面这个可以延伸。

6.  以延伸反应的产物为模板，以a/d为引物进行pcr。

7.  产物即为所需最终产物。
